We transiently transfected ER-negative HeLa cells with an expression vector for human ER[alpha] or ER[beta] along with an estrogen-inducible 3X-ERE-TATA-Luc firefly luciferase reporter plasmid and a constitutively active cytomegalovirus (CMV)-[beta]-galactosidase reporter plasmid and treated the cells for 24 hr with a solvent control or increasing concentrations of [E.
The resulting transfectants expressed generally increased, but variable levels of firefly luciferase from the construct containing the GA sequence, at a pH of 6.
