Dulbecco's modified Eagle's medium (DMEM), RPMI-1640, fetal calf serum (FCS), HEPES, L-glutamine, 2-mercaptoethanol, penicillin/streptomycin, bovine serum albumin (BSA), dinitrophenyl conjugated human serum albumin (DNP-HSA), p-nitrophenyl-N-acetyl-[beta]-D-glucopyranoside, and neutral red were obtained from Sigma Chemical Company (St. PRNT testing for WNV used a single basal medium Eagle--based overlay containing neutral red, while SLEV testing required application of a double overlay, the first without and the second with neutral red, applied 3 days after the first. Inhibition was quantitated by visual cytopathic effect (CPE), inhibition of the cellular uptake of the vital dye, neutral red (NR), and by virus yield reduction. |
