"If a patient is admitted with a suspected kidney injury, we can screen the injury using blood/urine with the help of an enzyme called
fumarase, and then a scan can pinpoint where in the tissue the injury is," says one of the researchers behind the study, PhD student from the Department of Clinical Medicine at Aarhus University, Per Mose Nielsen.
Fumaric aciduria is a rare congenital metabolic disorder characterized with increased fumaric acid which occurs as a result of deficiency of
fumarase which is one of the enzymes included in the tricarcoxylic acid (Krebs) cycle.
Proceeds from the fun day will be used to assist other families who have children with
fumarase deficiency or other serious diseases.

Four of 16 allozyme loci, creatine kinase (CK-B*),
fumarase (FH*), isocitrate dehydrogenase (ICDH-S*), and lactate dehydrogenase (LDH-B*), were unique in C.

We measured
fumarase activity by following the increase in absorbance at 240 nth at 25[degrees]C in a reaction mixture containing 30 mM phosphate (pH 7.4), 0.1 mM EDTA, and 5 mM L-malate (Racker 1950; Sigma Chemical Co., St.
(1986)]: acid phosphatase (ACP), alanine dehydrogenase (ALA), butyrate esterase (EST), [Beta]-hydroxybutyrate dehydrogenase (HBD), diaphorase (DIA),
fumarase (FUM), fructose-1,6-diphosphatase (F16), glucose-6-phosphate dehydrogenase (G-6), glutamic-oxalacetic transaminase (GOT), glyceraldehyde-3-phosphate dehydrogenase type 1 (GP1), isocitrate dehydrogenase (IDH), indophenol oxidase (IPO), leucine aminopeptidase (LAP), leucine tyrosine peptidase (PEP), malate dehydrogenase (MDH), malic enzyme (ME), phosphoglucose isomerase (PGI), phosphoglucomutase (PGM), 6-phosphogluconate dehydrogenase (6PGD), and shikimate dehydrogenase (SDH).
Amongst them, the
fumarase is considered as the fundamental metabolic switch regulating the metabolism of matter and energy (Kosaka et al., 2006).
The team applied a novel network algorithm to a published genome-scale model of human metabolism to design minimal "knockouts" for a wide variety of metabolic functions, such as phospholipid biosynthesis and the role of
fumarase in suppressing human cancer. The research suggests that the many pathways in the human metabolic network buffer each other to a striking degree, inducing "deep" epistasis -- the suppression of a mutation by one or more seemingly unrelated genes.
fumarase, fumarate reductase, citrate synthase were not isolated; however, aconitase and isocitrate dehydrogenase were isolated.
The 7-year-old, described by her mother as "the happiest little girl that you would ever meet," was diagnosed at age 2 with
fumarase deficiency.
