The restriction fragment patterns of these juveniles were identical to those of previously observed adult specimens, except for the two S.
Restriction enzymes seek a specific sequence, typically between four and eight bases in length, and cut the DNA at this location, creating discrete restriction fragments. Scientists can harness this phenomenon for genomic research.
A large-scale DNA isolation procedure (Dvorak et al., 1988) was used for the restriction fragment length polymorphism analysis (RFLP) and to optimize the amplification conditions for PCR markers.
Instead, restriction fragments of the putative hybrid nuclear PCR product were always the same size as those of the sea urchin.
Restriction fragments were separated by gel electrophoresis through 2 per cent TBE agarose gel (Genei, Bangalore).
The significant differences obtained in dynamic range did not depend on which spectral band was chosen for quantification among the 10 restriction fragments, except for a pair of 7mers coming from the H3 motif.
For the restriction digestion, 7.5 to 10 [micro]L of PCR products were used on the basis of expected numbers and sizes of restriction fragments. Restriction fragments were separated on 2.5% (w/v) agarose gels containing ethidium bromide, and were photographed under the Gel Doc 2000 (Bio-Rad, Hercules, CA).
