After 5 minutes incubation at room temperature, cobalt chloride solution was replaced with freshly prepared solution containing equal volumes of
ammonium molybdate solution (6.25%) and ammonium vanadate solution (0.42%).
100 g of seeds received, separately and/or combined, the application of 0.61 g peat inoculant based on Azospirillum brasilense; 0.61 g of peat inoculant based on Rhizobium tropici; and 0.07, 0.14, 0.21 and 0.28 g of
ammonium molybdate. These quantities were established based on the doses of 100 g of inoculant for 60000 seeds and 7.5, 15, 22.5 and 30 g Mo [ha.sup.-1].
A 0.68 M solution of
ammonium molybdate tetrahydrate (Sigma-Aldrich 277908) was prepared in a mixture of distilled water and dimethylformamide in a ratio of 5:1, respectively.
Reagents: 4 %
ammonium molybdate solution and 0,03 % hydrogen peroxide solution.
The color was developed with molybdate reaction which was as follows: The 500 L mixture (composed of 200 L of 2 %
ammonium molybdate and 300 L of 14 % ascorbic acid in 50 % trichloroacetic acid) was added to the above mixture (700 L) followed by the addition of 1 mL solution containing 2 % trisodium citrate and 2 % sodium arsinate in 2 % acetic acid to make the total volume of 2200 L.
