Serum diamine oxidase activity in patients with histamine intolerance.
The levels of TNF-a, IL-6, IL-1[sz], and diamine oxidase (DAO) were determined by enzyme-linked immunosorbent assay (ELISA) according to the manufacturer's instructions (R&D, USA), and the concentrations of cytokines were determined spectrophotometrically by the absorbance at 450 nm.
Diamine oxidase or DAO is responsible for breaking down dietary histamines and to a lesser extent, endogenous histamine.
Two oxime compounds, 1-(3-methylsulfanyl-[1,2,4]triazin-5-yl)- ethanone oxime (2a) and 1-(3-methylsulfanyl- [1,2,4]triazin-5-yl)-propan-1-one oxime (2b) displayed only a very weak effect on pea diamine oxidase. They were found to be non-competitive inhibitors characterized by high Ki values of 14 and 16 mM, respectively.
Diamine oxidase (DAO) level was measured in serum samples using a rat specific ELISA kit following manufacturer's instructions.
Stereochemistry of reactions catalyzed by L-lysine decarboxylase and diamine oxidase. Phytochemistry 18:771.
After histamine is methylated by HNMT, it is further metabolized by diamine oxidase or monoamine oxidase-B (MAO-B) (Maintz & Novak, 2001).
