The levels of NCG in rumen fluid were all around distance by methods of cation-exchange chromatography using NaOH as eluent and an IC column Cleanert IC-RP with Spec: 1 mL (250mmx4mm i.d.) as the partition column.

The extracted copper was stripped from the membrane disk using appropriate amounts of suitable eluent (the best eluent was 1 mol x [L.sup.-1] nitric acid).
- Launching an HPLC Column to Analyze Anions, Compatible with Hydroxide Eluents
Filtered and the filtrate washed with saturated dicarbonate solution three times, dry the organic with over anhydrous sodium sulfate overnight, filtered again, evaporated to dryness the filtrate and silica gel column chromatography (silica gel, 200-300 mesh; eluent, petroleum ether/ acetic acid acetate/methyl aclohol = 20 : 1 : 0.05) to get a reddish brown solid powder 0.377 g, yield 42.6%.
A good extraction solvent will contain all of the eluents in the chromatography process so that any reactions of the sample resulting in precipitates will occur prior to injection into the HPLC.
Eluent type and concentration also played an important role in SPE preconcentration system.
The solvent was evaporated under reduced pressure, and the product was purified by column chromatography on silica gel (eluent petroleum ether-ethyl acetate, 7:1).
