The reducing power (RP) of a compound may serve as a significant indicator of its potential antioxidant capacity to reduce the potassium ferricyanide ([Fe.sup.3+]), to form potassium ferrocyanide ([Fe.sup.2+]).
These authors revealed that upon sunlight exposure, the conversion of ferricyanide to ferrocyanide and the reverse reaction depended strongly on the pH of the solution whether or not the Ti[O.sub.2] catalyst was present.
This assay was used to convert the potassium ferricyanide complex to potassium ferrocyanide complex, which would constitute ferrous complex by reducing ferric chloride [34].
After washing with PBS, cells were incubated at 37[degrees]C with freshly prepared senescence-associated SA-[beta]-Gal staining solution: 1 mg/ml 5-bromo-4-chloro-3-indolyl P3-D-galactoside (X-Gal), 40 mM citric acid/sodium phosphate, pH 6.0, 5 mM potassium ferrocyanide, 5 mM potassium ferricyanide, 150 mM NaCl, and 2 mM MgCl2.
Sodium dihydrogen phosphate (NaH2PO4), potassium ferrocyanide ([K.sub.3]Fe[(CN).sub.6]), trichloroacetic acid (TCA), iron-III-chloride (Fe[Cl.sub.3]), potassium peroxydisulfate ([K.sub.2][O.sub.8][S.sub.2]) copper-II-chloride (Cu[Cl.sub.2]), sodium acetate (NaCH3COO), hydrochloric acid (HCl), tris, iron-II-sulfate (FeS[O.sub.4]), iron-II-chloride (Fe[Cl.sub.2]), disodium hydrogen phosphate ([Na.sub.2]HP[O.sub.4]), methionine, ethanol, ethylene-diaminetetraacetic acid (EDTA), ammonium thiocyanate (N[H.sub.4]SCN), sodium bicarbonate (NaHC[O.sub.3]), sodium hydroxide (NaOH), disodium sulfate ([Na.sub.2]S[O.sub.4]), sodium perchlorate (NaCl[O.sub.4]), and disodium carbonate ([Na.sub.2]C[O.sub.3]) were obtained from Merck (Merck, made in Germany).
The supernatant collected was assayed for ferrocyanide content using 4, 7-diphenyl-1, 10-phenanthroline disulfonic acid disodium salt, absorption was recorded at 535 nm ([member of] = 20,500 [M.sup.-1] [cm.sup.-1]).
