This method, based on eDNA representational difference analysis (cDNA RDA), uses 96 hexanucleotides that are not suitable for priming ribosomal RNAs but that normally prime most of the genome of an RNA virus as primers for reverse transcription in eDNA RDA (1). Where there were choices as to which method to apply, the small-subunit ribosomal RNA gene was considered to be the optimum genetic locus to target. This technique scrutinizes a specific segment of the microbes' genomes, called 16S ribosomal DNA, that tends to vary from species to species but is similar in closely related species. |
