In pilot-controller terminology, it means, “Operate the radar beacon transponder on [the designated code] in mode A/3.”

squawk

A code meaning, “Switch the IFF (identification friend or foe) master control to the ‘normal' (mode and code as directed) position.” In some air forces, the term sing canary is used.
