Knockdown

the condition of a boxer after a blow in which he is judged by the referee to be unable to continue the match for several seconds. The command to resume the match is given only after a count of eight, and only if the athlete is able to continue. In personal championship fights in the USSR, the match is ended after the second knockdown.

For the second siRNA transfected (siRNA2), as shown in Figure 4C, small decrease in CYP19 gene transcript abundance was seen when transfected with 100nM siRNA (siRNA2) concentration, however, percentage gene knockdown achieved was only about 40 percent (Figure 4B).
Further studies may determine the longevity of gene knockdown and whether the effect could be incorporated into progeny or enhanced by multiple applications.
This delivery strategy produces over 500-fold increase in efficiency in mice compared to injection of cholesterol-siRNA alone and is the first delivery system to demonstrate cholesterol-siRNA mediated gene knockdown in monkeys.
Data from new studies in mice and non-human primates show high levels of target gene knockdown at very low doses with no toxicity.
First, these results demonstrate the highly robust level of target gene knockdown and attractive pharmacologic profile achievable with RNAi therapeutics.
As a GalNAc-siRNA conjugate, ALN-AT3 achieves target gene knockdown with subcutaneous dose administration, a highly preferred mode of administration in the setting of hemophilia.
Importantly, DPCs display a low toxicity profile enabling siRNA redosing and long-term target gene knockdown.
