Caption: FIGURE 1: Change in emission spectrum of B (1 [micro]M) on addition of TNP (150 [micro]M) in THF: HEPES (9.5: 0.5) (]/]).
The reagents used for NPs synthesis were obtained from Sigma-Aldrich: HAu[Cl.sub.4]x3[H.sub.2]O, HEPES, N[H.sub.2]OHxHCl, and sodium citrate.
KCl, 140; MgCl [sub]2 , 1; 10 [micro]M nigericin; buffered with one of the following organic buffers: 20 mM 2-(N-morpholino) ethanesulfonic acid (MES, pH 5.5), 20 mM HEPES (pH 7.5) or 20 mM 3-(cyclohexylamino)-2-hydroxy-1-propane-sulphonic acid (CAPSO, pH 9.5), and were adjusted (37[degrees]C) to the correct pH with 4N NaOH.
pH was adjusted to normal physiological range of 7.35-7.40 with NaHC[O.sub.3] in all groups except for the HEPES 1 and HEPES 2 groups (table 1).
Increasing the concentration of NaCl from 0.01M to 0.3M in 50mM HEPES pH7, 50mM L-Arg & Glu, 0.1M trehalose increased [B.sub.22] value from around zero to +6.6 x [10.sup.-4] mol x mL x [g.sup.-2] (Figure 8).
A6909; Sigma) was diluted with 0.01[micro]mol/L HEPES buffer (pH 7.4) to the final desired concentration.
EndoIV reactions consisted of 100 mM Hepes, pH 7.5; 100 mM KCl; 20% glycerol; 0.02% TX-100; 1 ng (3.2 nM, assuming a molecular weight of 31,480 g/mol) EndolV, and 100 [micro]M of the indicated metals in 10 [micro]L.
To search for dynactin-dynein motor molecules in Spraguea sporoplasms, purified spores were incubated in 0.1 M HEPES buffer (pH 7.0) and transferred to glass coverslips.
Dulbecco's modified Eagle's medium (DMEM), RPMI-1640, fetal calf serum (FCS), HEPES, L-glutamine, 2-mercaptoethanol, penicillin/streptomycin, bovine serum albumin (BSA), dinitrophenyl conjugated human serum albumin (DNP-HSA), p-nitrophenyl-N-acetyl-[beta]-D-glucopyranoside, and neutral red were obtained from Sigma Chemical Company (St.
To initiate spore discharge, spores of Spraguea lophii were incubated in 0.1 M HEPES buffer at pH 7.0 (with 50 nM [Ca.sup.++]) for 1 h.
After the HAEC were treated with various concentrations of [As.sup.3+] for 14 hr, they were incubated for 20 rain in HEPES buffer (pH 7.4): 140 mM NaCl, 2.7 mM KCl, 1.0 mM Mg[Cl.sub.2], 5.0 mM glucose, 25 mM HEPES, and 3 [micro]Ci/mL [[sup.3]H]-L-arginine.
