Our approach to DNA sequencing by primer walking bypasses the requirement for custom primer synthesis and relies on a universal set of 256 presynthesized oligonucleotide adaptors, referred to as indexers (17), and type IIS restriction endonucleases that cleave DNA at defined distances from their recognition sites, generating ambiguous 4-nucleotide (nt) 5' overhangs (18).
To further characterize the phylogenetic relationships between the CHIKV strains in this outbreak with strains previously found in Thailand and neighboring regions, we performed full-length viral genomic sequencing from srains from 4 samples by primer walking (4).
