This agglutination is likely due to spontaneous
acrosome reactions which could have occurred at some point during freezing or thawing.
A stimulatory effect of methyl xanthine on capacitation and
acrosome reaction has also been demonstrated (Aitken et al., 1993).

The reason for this is a membrane protective capacity of still unknown egg yolk components (Witte et al., 2009), but the low-density lipoprotein fraction (LDF) of egg yolk (Awad and Graham, 2002) may interact with the plasma membrane of sperm and also decrease the efflux of cholesterol and phospholipids from the sperm membrane and thus prevent premature capacitation and subsequent
acrosome reaction (reviewed by Purdy, 2006).
Contact with the vitelline layer overlying the microvilli initiates the typical
acrosome reaction, in which the outer acrosomal membrane and plasma membrane fuse at the apex and roll back in a manner that releases the contents of the acrosomal vesicle onto the vitelline layer [ILLUSTRATION FOR FIGURES 25, 26 OMITTED].
Indeed, SA certainly does not evaluate many of the parameters of sperm that may impact MFI such as an intact
acrosome reaction [8].
These are suitable conditions for sperm cell capacitation 10 and also to evaluate their ability to undergo
acrosome reaction. The latter was induced by incubating 5x106 sperm cells with 20 [micro]L of perivitelline layer (PVL) and kept for 40 min at 37 [degrees]C (Ahammad et al., 2011b).
