The product (ivosidenib) is an isocitrate
dehydrogenase-1 (IDH1) inhibitor indicated in the United States as a treatment for acute myeloid leukaemia with a susceptible IDH1 mutation under the conditions where in patients with newly diagnosed IDH1-susceptible acute myeloid leukaemia who are up to 75 years old or have comorbidities that preclude the use of intensive induction chemotherapy and in adult patients with R/R AML with a susceptible IDH1 mutation as detected by an FDA-approved test.
In addition, significant decreases were observed in the activities of pyruvate dehydrogenase complex (-41%), isocitrate
dehydrogenase (-27%), and [alpha]-ketoglutarate dehydrogenase complex (-57%).
IDHs are homodimeric enzymes in the Krebs cycle that normally catalyze the oxidative carboxylation of isocitrate
to [alpha]-ketoglutarate ([alpha]-KG; Figure 6).
Since 2012, histopathological markers such as MGMT, isocitrate
