Neurofibril

 

a microscopic filament that is found upon treatment of nerve cells—neurons—and the longest outgrowths of the nerve cells—axons—with silver salts and other reagents.

In the late 19th and early 20th centuries, neurofibrils were ascribed the function of conducting nerve impulses. This view proved to be erroneous when it was found that nerve impulses are conducted by the external membrane of the neuron. Electron microscopy revealed two kinds of longitudinal neurofibrils in the outgrowths of neurons: neurotubules and neurofilaments. Neurotubules are from 20 to 25 nm in diameter and are formed from the protein tubulin; they are believed to transport substances along the axon. The threadlike neurofilaments are formed from a protein similar to the muscle protein actin. Neurofilaments are especially numerous in the movable terminal portions of growing axons.

Comprobando el papel especifico de la apoE en la formacion de placas amiloides y enmaranados neurofibrilares, existen estudios que muestran que la apoE es uno de los componentes de las placas amiloides cerebrales.
Desde que se descubrio la proteina Tau como componente llave en los enmaranados neurofibrilares (ENF) en la EA, esta proteina paso a ser objeto de intensas investigaciones.
A traves de los biomarcadores, en los pacientes con EA se observa una disminucion de la sintesis de alipotroteina E , una mayor cantidad de Beta-amiloide en las placas y un aumento de los enmaranados neurofibrilares agregando las proteinas Tau.
Tambien, se ha establecido que la excitotoxicidad medida por el receptor NMDA aumenta la fosforilacion de la protina tau, y por lo tanto, puede estar implicada en la degeneracion neurofibrilar (130).
Por lo que respecta a los correlatos histopatologicos del envejecimiento normal, en la actualidad persiste la controversia acerca de si los hallazgos basicos de la EA --los ovillos neurofibrilares (ONF) y las placas seniles (PS)-- son especificos de la enfermedad o pueden aparecer tambien en el cerebro de ancianos sanos.
