The main sulphur-compound is S- benzyl phenylmethane thiosulfinate (called Petivericine) and other isolates that include s-benzyl (2-hydroxyl) ethane thiosulfinate, S - (2 - hydroxyethyl) phenylmethane thiosulfinate, and S- (2-hydroxyethy) 2- (hydroxyethane) thio sulfinate [8,9].
Whole cell lysates were extracted using EBC lysis buffer [120 mM NaCl, 0.5% Nonidet P-40, 5 [micro]g/mL leupeptin, 10 [micro]g/mL aprotinin, 50 [micro]g/mL phenylmethane sulfonyl fluoride, 0.2 mM sodium orthovanadate, 100 mM NaF, and 50 mM Tris-Cl (pH 8.0)] and isolated by centrifugation at 12,000 xg for 10 min at 4[degrees]C.
Brain regions (OE, OB, cerebral cortex, cerebellum) were homogenized by a motor-driven pestle on ice in 1x RIPA buffer (Millipore) supplemented with 1 mM phenylmethane sulfonyl fluoride (PMSF), 1 mM [Na.sub.3]V[O.sub.2], 10 mM NaF, phosphatase inhibitor cocktails (Sigma), and Roche Complete Mini EDTA-free Protease Inhibitor Cocktail Tablet (Roche).
Immobilized pH gradient (IPG) strips (pH 3.0 to 10.0 non-linear (NL); 17 cm), urea, thiourea, dithiothreitol (DTT), 3-([3-cholamidopropyl] dimethyl-ammonio)-1-propane sulfonate (CHAPS), ampharmalyte pH 3 to 10, phenylmethane sulfonyl fluoride (PMSF), iodoacetamide (IAM) were purchased from Bio-Rad (Richmond, CA, USA).
Gastrocnemius was homogenized in RIPA buffer (10 [micro]L/mg) with protease inhibitors cocktail, sodium deoxycholate 12.5 mM, phenylmethane sulfonyl fluoride 100 mM, sodium orthovanadate 12.5 mM, and with phosphatase inhibitors (Sigma-Aldrich, Madrid, Spain) for pAkt/Akt and pFoxO1/FoxO1.
Pieces of muscle weighing 1.0 g were added to 9.0 ml of ice-cold 20 mM Tris buffer including 1mM phenylmethane sulfonyl fluoride, 3 [micro]g/ml leupeptin and 1 mM EDTA, and homogenized with a Polytron on ice for 30 s two times with a 30-s interval.
