The rationale for the assay is that a contact activator is used to provide a surface for high-molecular-weight kininogen, prekallikrein, and FXII, thereby stimulating the production of FXIIa.
During pre-incubation time, contact factors (XII and XI) are activated in the presence of high molecular weight kininogen and prekallikrein. The activated factor XI, converts factor IX to [IX.sub.a].
During normal contact activation (caused by tissue damage), activated factor 12 cleaves prekallikrein to kallikrein which then leads to bradykinin formation.
The kallikrein-kinin cascade is stimulated as a result of activated kallikrein (or Hageman factor/Factor XII and prekallikrein), or microbial proteases cleave off bradykinin directly from either high-molecular-weight or low-molecular-weight kininogen in the plasma (3-11,13) which causes pain and extravasation.
Plasma levels of factor XII, prekallikrein and high molecular weight kininogen in normal blood donors and patients having suffered venous thrombosis.
